Then your test is wrong. The shortest valid line is
n,1,2,3,4,5\n
But that's not the way to go anyway.
The current program does nothing useful so I have to speculate on how
you will eventually do some real processing. The simplest way to know
if you have exactly five numbers is to parse all the numbers and report
when you don't get five.

No, you can the buffer until index == 0 and test !isdigit(buffer[0]).
You find that 'a' is not a digit and do index--. index is an unsigned
int type so the subtraction wraps round and index will have a value of
something like 18446744073709551615 (that's what it is on my system).
Your program that tests !isdigit(buffer[18446744073709551615]). What do
you think it will find?
On my system, gcc has an -fsanitize=undefined option that does a lot of
run-time checking. Running you program with the input in question gives
me this helpful exchange:
$ ./a1 /dev/stdin ✘
abc,def,ghi
*****************************

But that does not answer my question. I know you will eventually parse
the numbers -- that's why I asked about the test above. If you write
that program correctly you won't care about the non-printing characters.
