It keeps your custom setting as long as you don't close that window /
tab. As soon as you open another window / tab, it returns to the deaful
option.

Not within Safari itself.
I don't know if one of the system utility apps like TinkerTool or Onyx
can change it more easily ... and make it easier to revert if things go
wrong!
It's not really something you should probably be playing with and I
don't know if it still works, but you used to be able to change it
using a Terminal command:
defaults write com.apple.Safari CustomUserAgent "\"useragentstring\""
where you replace useragentstring with the one you want. The easiest
way to find that string is to change the User Agent in Safari to the
one you want, then go to the Develop > User Agent > Other and copy the
text from that window.
