It depends what you mean by |{x}| = 1. Possibly you mean
a) x occurs only once in {x};
or possibly you mean
b) x is a member of {x} but no other object is.
Now (b) can be defined using the usual set axioms and relations.
But (a) can only be defined if you add an extra axiom and add an extra
relation to the usual two (which are the = and IsMemberOf relations).
