You have squared it already -- you can't square it again.

You have 6.45 * 10^8 u^2 / inch^2

Your spot is 100 x 100 u = 10^4 u^2

So you get 6.45 * 10^4 spots / in^2

Each spot is 24 bits or 3 Bytes.
So you get just under 200 kBytes / in^2
That would be an appallingly low density, even if you could do it.
The problems involved in reading the color quickly, cheaply, and 
consistently would not be trivial.
