In Yosemite, is there a one-step way to click on the Desktop
folder in the Dock and have it open as a Finder window, or do
I always have to click the folder once and then click on the
Open in Finder option? I haven't been able to find any settings
either in the folder options or Finder preferences to do this
with a single click.

You could always create an AppleScript / Automator app that opens the
window and put that in the Dock instead of the Desktop folder.

tell application "Finder" to open the (path to the desktop folder)

the following keyboard shortcut: CMD + Shift + D ('d' as in Desktop) 
opens the Desktop in a new finder window.

Also, I just found myself having to open the Downloads folder, so it's
easier for me to just remember one set of keystrokes to accompany my
trackpad action that I can use for any window. But thanks to CB for the
shortcut.

I just click on the downloads folder in the Dock.

Michelle, what did you do to your Finder preferences such that a single
click opens the Downloads folder into a Finder window? That's the solution
I've been after during this whole discussion! I thought it had disappeared
from recent versions of OS X.

I misunderstood you.  But you can do it with two clicks; right-click
(or two-finger click with a trackpad) on the Downloads folder in the
Dock, and choose the Open item at the bottom of the popup menu.

Some other similar shortcuts when in the Finder (and Open and Save
windows) are:

    Command + Shift + A    for the Applications folder
    Command + Shift + U    for the Utilities folder
    Command + Shift + H    for your user's Home folder
    Command + Shift + C    for the "Computer" folder (mounted drives)

although some don't work in some older versions of Mac OS X.

Why bother with three keys when one (or two) will do? 
Set-up an AppleScript / Automator app and assign it to a function key.
I haven't checked, but you may even simply be able to assign
Command-Shift-D to a function key.

I haven't bothered under OS X, but in OS 8 and 9 I had the function
keys set to run various internet applications, so whenever I logged on
via my dial-up account I could press F1, F2, F3 and have the email,
newsreader, and web browser applications all open by the time the modem
had logged in. I can't remember what F4 is set-up to do, but F5 opened
up my "Downloads" folder in Finder. :-)
